Ben Olsen and Clyde Simms scored in the 21st and 55th minutes, respectively, for D.C. United, while Sergio Santana had the only goal for CD Guadalajara in the 60th minute.
This is also the first victory by a USA club at the Copa Nissan Sudamericana since CONCACAF teams began competing in 2005.
The second-leg of Series 5 between CD Guadalajara and D.C. United will take place at the Estadio Jalisco in Guadalajara, Mexico on 2 October.
The 2007 Copa Nissan Sudamericana continues for the other CONCACAF teams next week as Club América (MEX) holds a 4:1 advantage in Series 8 when the face the defending champion Pachuca CF (MEX) in the second-leg at the Estadio Azteca in Ciudad de México on 3 October.
